A Two-Stage, High-Cell Density Process for Heterotrophic Omega-3-Enriched Algal Production Using Crude Glycerol and Integrated within the Biodiesel Refinery Disclosed is a two-stage, integrated, high-cell density fermentation process for production of omega-3-rich micro-algae which could then be used directly as a food and/or animal feed or further processed for isolation of the DHA or omega-3 fatty acids for nutraceutical and fuel use. The micro-algae used in the fermentation process are Schizochytrium limacinum SR 21, a known high-cell density species with unique abilities for efficient metabolism of glycerol carbon sources. A key to the process is the developed two-stage process using two separate reactors that takes advantage of the algae’s unique growth processes by controlling operating conditions in a first stage focused on cell number growth and then shifting operating conditions to induce a second stage focused on cell mass and fatty acid accumulation. The second stage occurring in the second reactor operates at two different operating conditions during its fed batch cycle so as to induce, at the end of the cyle, a higher percentage of DHA and/or omega-3 fatty acids in the fatty acid profile. Operating conditions that are controlled and shifted to induce maximum production in the two stages are carbon and nutrient feed types and concentrations, dissolved oxygen content, and temperature. The result of the combined two-stage process is a five-day fermentation regime that allows for extremely high cell densities (~150 g/L) and high productivity (~1.2 g/L hr) of algae with total, PUFA and DHA fat percentages of ~50, 25 and 20%, respectively. The entire process is efficiently integrated within the biodiesel refinery process. The main feedstock utilized by the micro-algae and for which the whole process and choice of particular organism is designed is crude glycerol produced as a by-product of the biodiesel refining process. After some pre-treatment which can be performed on-site, the crude glycerol is used as a carbon source to ferment the algae. The produced algae can then be put through a downstream separation process whereby the algae biomass is directly treated to produce: (1) omega-3 fatty acids for nutraceutical sale; (2) biodiesel from the non-omega-3 fatty acids; (3) crude glycerol that is reclaimed as carbon feed for future fermentation and algae production; and (4) algal biomass waste which can be used as an animal feed both internally in the fermentation process or externally.

Disclosed is a novel integrated nutrient recovery system that works in series with an on-farm anaerobic digester for removal and recovery of nitrogen (N) and phosphorous (P) in the form of ammonia sulfate solution and phosphorous-rich solids, respectively. The phosphorous-rich solids are obtained through dosing of specialized flocculants and subsequent settling and dewatering. Ammonia air stripping using a closed-loop recycle with novel tower and packing design with subsequent acid treatment for ammonia salt recovery is used for the ammonia recovery. Control of the effluent pH after ammonia treatment is accomplished by absorption of acid components in biogas that is bubbled through the effluent. The result is a pH controlled effluent and a scrubbed biogas with complete removal of hydrogen sulfide, partial removal of carbon dioxide and an associated increase in methane concentration. Placement of the unit operations is flexible within the integrated system allowing for added flexibility in operation for optimization with various types of anaerobic digested (AD) manure effluent.
